Effect of Tunic and Plant Growth Regulators on Dormancy Breaking and Growth of Gladiolus Cormel




Abstract
This study was performed to investigate the effects of tunic and plant growth regulators (PGRs) on dormancy breaking and growth of Gladiolus cormel, which obtained from cormlet produced in vitro. The numbers of tunic were 3 in ¢¥Nova Lux¢¥ and 6 in ¢¥Spit & Span¢¥ by the naked eyes. Sprouting percentage of the cormels was 90% in ¢¥Nova Lux¢¥ when the tunic was removed only one, whereas that of ¢¥Spic & Span¢¥ was 80% by the removal of 5 tunics. Sprouting percentage was 0% in ¢¥Nova Lux¢¥ and ¢¥Spic & Span¢¥ when tunic was not removed, and also the percentage was below 20% when PGRs was not treated as tunics were removed. But, in combination treatment of the tunic removal and kinetin, GA©ý, TDZ, BA, and BA+GA©ý dipping, the percentage was over 80% except kinetin treatment. Especially, in ¢¥Nova Lux¢¥ dipping treatment in BA and BA + GA, solution was 100% in sprouting and the days to sprouting were largely shortened regardless of temperature. ¢¥Spic & Span¢¥ was also similar in tendency. In TDZ treatment sprouting was abnormal morphologically and died after sprouting. In the case of dormancy-broken cormels, growth of ¢¥Nova Lux¢¥ corm decreased by tunic removal and PGRs treatments, while that of ¢¥Spit & Span¢¥ corm were promoted and generated many of new cormel.
